Monitoring the Expression of the HLA-A2.1 Transgene

The strain C57BL/6-TgN(HLA-A2.1)1Enge (Stock #003475) has recently been made available for sale by The Jackson Laboratory. We are distributing these mice as hemizygotes (Tg/-) while we are establishing a stable, homozygous colony. Transgenic mice will be identified by monitoring the expression of the HLA-A2.1 transgene by flow cytometry prior to distribution. In addition to identifying mice carrying the transgene, such flow cytometric analyses will ensure that the transgene is expressed.
Preliminary quantitative analyses of the transgene in our colony indicate that the transgene copy number in this stock may be varying, either due to multiple integration sites or locus instability. In the future, we hope to be able to provide more definitive information about the transgene copy number and/or insertion sites in this stock.
