| This strain carries a transgene that directs skin-targeted overexpression of the human DNA repair protein MGMT (O6-methylguanine-DNA methyltransferase). The transgenic strain exhibits a significantly reduced incidence of alkylation-induced skin tumors. It may be used (1) to investigate the role of the MGMT DNA repair enzyme in protecting cells from premutagenic and precarcinogenic O6-alkylguanine lesions, (2) as an indicator system for O6-alkylguanine generating carcinogens (e.g. N-nitrosamines, N-nitrosoureas) and (3) to explore the pathophysiological significance of these lesions for various end points. Moreover, skin tumors induced on these transgenic mice may be used as a model for chemotherapy resistant tumors expressing high levels of MGMT. | ||
