| Mice homozygous for this Oct4-EGFP mutation are viable and fertile. They harbor an IRES-EGFP fusion cassette downstream of the stop codon of the Oct4 (Pou5f1) gene. When treated with specific transcription factors (Oct4, Sox2, c-Myc and Klf4), some Oct4-EGFP murine embryonic fibroblasts (MEFs) have the properties of induced pluripotent stem (iPS) cells. Such iPS cells have the DNA methylation, gene expression and chromatin state of embryonic stem cells and can form viable chimeras, contribute to the germ line, and generate live late-term embryos when injected into tetraploid blastocysts. These Oct4-EGFP mutant mice may be useful for fluorescent labeling of embryonic stem cells, as well as for the selection of iPS cells (i.e. epigenetic reprogramming of somatic cells into pluripotent embryonic stem cells).

| The donating investigators report an inducible binary transactivation system that permits the control of transgene expression in a tissue-specific and inducible fashion in mice. Transcription of a target transgene is kept silent until turned on by the application of an exogenous compound. No phenotypic defects are involved. The components of the construct used includes insulator DNA segments (5' region of chicken beta globin gene) flanking both ends of the GLVP transactivator which is driven by the transthyretin enhancer/promoter. The GLVP portion consists of a mutated human progesterone receptor ligand binding domain fused to the yeast GAL4 DNA binding domain and the HSV VP16 transcriptional activation domain. In the published reference, donating investigators crossed the GLVP transactivator mouse with a second target mouse line (17x4-tk-hGH). New Strains Under DevelopmentThe Jackson Laboratory serves as a worldwide distributor and national repository for common and rare strains of inbred mice and mice carrying spontaneous mutations or induced mutations (i.e., transgenic, targeted/"knockout", or chemically induced mutations). At any one time, we have over 100 strains at various stages of development and colony expansion. Strains "Under Development" fall into two categories depending on the anticipated demand from the scientific community.
- Receive periodic updates on the status of the colony UNDER DEVELOPMENT
- Obtain advance notification of strain availability and opportunity to order prior to the strain being published as available
- Provide input affecting speed and quantity of availability
It is VERY IMPORTANT that you register interest in strains Under Development. The anticipated demand for a strain enables us to determine effectively the distribution plan for each strain Under Development. Registering interest also provides benefits to you (including advance notification of pending availability). Whether a strain is made available from a live colony OR from our cryopreservation repository, you may want to consider the option of Dedicated Supply.
