Himalayan, an allele at the tyrosinase locus, arose spontaneously in the offspring of a cross between a DBA/2J male and an AKR/J female at The Jackson Laboratory in 1958. The first affected, a male, was backcrossed to an AKR/J female and the Tyrc-h/Tyrc carriers were mated to a C57BL/10Gn. The pairs were mated to produce the homozygous Tyrc-h/Tyrc-h mouse. The stock was then maintained by mating sibling homozygotes to make the inbred strain a/a Tyrc-h/Tyrc-h. It was cryopreserved in 1998 by mating homozygotes at F140.

| General Note | Tyrc-h, himalayan. The himalayan mutation appeared in the offspring of a cross between the DBA/2 and AKR/J strains at The Jackson Laboratory. In homozygotes, the first coat is a uniform light tan. At the first molt, the body hair becomes lighter and the ears, nose, tail, and scrotum become dark as in Siamese cats. Body hair may be very light or quite sooty, depending on the genetic background. Eyes are slightly pigmented and appear red. Tyrc-h/Tyrc and Tyrc-h/Tyrc-e mice are intermediate between the homozygotes (J:13075). Melanin granules of the retina are indistinguishable from those of chinchilla or of wild-type homozygotes (J:5001). Pigment synthesis in himalayan mice is temperature-sensitive; pigment develops in growing hairs of mice housed at 15oC but not in those housed at 30oC (J:6275). In explanation of this effect, it has been shown that tyrosinase in these mice is heat-labile (J:12173), being maximally active at well below body temperature.Tyrosinase of Tyrch homozygotes is strongly bound by an inhibitor at 37oC but not at lower temperatures (J:6508). The mutation in the himalayan allele was found to be an A to G point mutation at nucleotide 1259 that results in an amino acid change at position 420 from histidine to arginine (J:9824). | ||

| Molecular Note | Characterization of this allele shows an insertion of DNA comprised of a 5.5kb virus-like element, VL30, into the first intron of the agouti gene. The VL30 element itself contains an additional 5.5 kb sequence, flanked by 526 bp of direct repeats. The host intergration site is the same as for at-2Gso and Aw-38J and includes a duplication of four nucleotides of host DNA and a deletion of 2 bp from the end of each repeat. Northern analysis of mRNA from skin of homozygotes shows a smaller agouti message and levels 8 fold lower than found in wild-type. [MGI Ref ID J:16984] [MGI Ref ID J:24934] | ||

Cryorecovery - Standard. The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery. Cryorecovery to establish a Dedicated Supply for greater quantities of mice. |
