Strain Development
Mahoganoid (Mgrn1md) arose spontaneously on strain C3H/HeJ at the Jackson Laboratory in 1957. The Igl-1b allele at the immunoglobulin lamba chain cluster 1 locus is found in strains SJL, YBR, BSVS and others and the Igl1-rlo allele occurs in strains SJL, BSVS and others. In 1972 Dr. M. C. Green received the fused phalanges (Fbn2fp) stock from Dr. Katrina Hummel. Fused phalanges arose spontaneously in strain C3HeB/FeHu. The stock Dr. Green received had been crossed once to C57BL/6J and it was then brother x sister bred to F20 in 1976. At F18 the stock was given to Dr. Muriel Davisson and it was continued to be inbred to F21 when a fused phalanges male was outcrossed to a homozygous mahoganoid (Mgrn1md) female. This mahoganoid female was the N2 offspring of a backcross that began with a mating of C57BL/6J x C3H/He-Mgrn1md/+ from which an F1 was bred back to a C3H/HeJ-Mgrn1md/Mgrn1md, which was at F69. The N2 homozygous female resulting from this breeding was bred with the F21 fused phalanges male and their offspring were bred brother x sister to F20. At that point, a mahoganoid male was outcrossed to an Lystbg homozygous female on a congenic SJL/J background at generation N12F4 from Dr. John Roths. The resulting stock was sibling mated and a homozygous mahoganoid female was backcrossed to an SJL/J male. The stock was then bred as homozygous for mahoganoid (Mgrn1md),Igl-1b and Igl1-rlo all on chromosome 16. It was cryopreserved in 1991 by mating homozygotes at F27 and F28.

General Note The original md mutation arose in the C3H/HeJ strain. This gene is similar to mahogany (Atrnmg) in its effects on coat color, but causes less darkening of the back, ears, and tail of agouti mice (J:13379)(J:13791).
Molecular Note The mutation in md has been attributed to a 5 kb retroviral insertion located in the intronic region 5' of exon 12. Northern analysis and RT-PCR indicate that the expression of transcript is greatly reduced in these mice. [MGI Ref ID J:80182]

Supply Notes Cryorecovery - Standard.
The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery.
