The transgenic construct for Tg(Pfkl)224Yg has 2.9 kb of the 5' genomic region, the first exon, first intron, and some of the second intron of Pfkl followed by the cDNA sequence extending to the 3' untranslated sequence and then genomic sequence from the 3' untranslated region was place downstream of this. A 22 base pair deletion was introduced into this 3' untranslated sequence in order to provide a system for detection of transgene-derived transcript. This construct was microinjected into fertilized eggs from a CByB6F1 female crossed with a B6D2F1 male and these were implanted into CD1 mothers. Line 224 was found to have several dozen copies of the transgene. In 1994 homozygous males and females from line 244 were sent from Dr. Yorum Groner at The Weizmann Institute in Rehovot, Israel to The Jackson Laboratory. This strain was then maintained at The Jackson Laboratory by sibling breeding homozygotes and in 1995 homozygous embryos were cryopreserved.
