Former Names B6SJL-Tg(BARK12)12Wjk/J (Changed: 04-MAY-07 ) B6SJL-TgN(BARK12)12Wjk (Changed: 15-DEC-04 ) B6SJL-TgN(BARK12)12Wjk/J (Changed: 15-DEC-04 ) Type Mutant Strain; Transgenic; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse Generation N8 Donating Investigator Walter Koch, Duke University Medical Center Description
Mice carrying this transgene show a 3 to 5 fold overexpression of bovine beta-adrenergic receptor kinase-1. Myocardial beta-adrenergic receptors are hyperphosphorylated and desensitized in hemizygotes resulting in attenuated cardiac function in response to beta-agonists.

assigned by genotype
Tg(Myh6-ADRBK1)12Wjk/?
involves: C57BL/6 * SJL
- cardiovascular system phenotype
- abnormal heart rate (MGI Ref ID J:37034)
- heart rate failed to increase in response to the beta-agonist isoproterenol
- rises in pressure induced by the beta-agonist isoproterenol are depressed in these mice by about 20% at the highest dose
- increased left ventricle diastolic pressure (MGI Ref ID J:37034)
- there is a blunted response to rises in pressure induced by the beta-agonist isoproterenol
- rises in pressure induced by the beta-agonist isoproterenol are depressed in these mice by about 20% at the highest dose

| Promoter | Myh6, myosin, heavy polypeptide 6, cardiac muscle, alpha, mouse, laboratory | ||
| Molecular Note | The entire bovine adrenergic receptor kinase, beta 1 (ADRBK1) coding region was ligated to a 5.5 kb fragment of the murine adult myosin, heavy polypeptide 6, cardiac muscle, alpha (Myh6) promoter. Overexpression of the transgenic protein was detected by Western blot analysis of heart extracts. Biochemical assays showed that kinase activity was approximately 3 times greater in heart extracts from transgenic mice than from wild-type mice. The Myh6 promoter drives expression in myocardial tissue. [MGI Ref ID J:37034] | ||
