Generation DefinitionsDonating Investigator Klaus Rajewsky, Max-Delbruck-Ctr. for Molecular Medicine Description
These transgenic mice carry the Cre recombinase gene under the control of the rat nestin promoter. When crossed with a strain carrying a gene flanked by loxP sites, the flanked gene will be removed in cells expressing cre, generating a proportion of mosaic animals. All adult organs, including germ-line cells, may be affected. The balancer2 line of transgenic mice show greater variability and instability in their generation of mosaics than the balancer1 line (Stock No. 002858). This variability of the balancer2 line can be of particular benefit if the gene of interest is vital and mosaic individuals are used to bypass lethality.Development
The transgene contains the gene for Cre recombinase under the control of the rat nestin promoter and second intron enhancer. The "NesI2Crec-1" construct was used. B6CBAF2 embryos were injected. The strain is currently on a mixed B6,129,CBA background.

| Site of Expression | induces variable mosaicism/partial deletion of loxP-flanked alleles before embryonic day 10.5 and in all adult organs (including germline), producing mosaic mice consisting of cells that are either wildtype or mutant for the gene of interest, more variable mosaicism than the "balancer 1" line (Stock No. 002858) | ||
| Expressed Gene | cre, cre recombinase, bacteriophage P1 | ||
| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||

| Driver Note | Nes | ||
| General Note | Mice carrying this "balancer 2" transgene show greater variability and instability in their generation of mosaics than do transgenic mice carrying the "balancer 1" transgene. | ||
| Molecular Note | The transgenic construct contained a nestin promoter followed by cre recombinase sequence adjoined to a nuclear localization signal. Intron 2 of the nestin gene was included downstream of a polyadenylation signal. While the endogenous nestin promoter is CNS-specific, Southern blot analysis showed the ubiquitous expression of cre in transgenic mice. Variable mosaicism/partial deletion of the loxP-flanked allele occurs, and it may be detected in all adult organs including germ line cells. [MGI Ref ID J:67926] | ||

Animals Provided
Price (US dollars $) Cryorecovery* $3000.00 At least two mice that carry the mutation (if it is a mutant strain) will be provided. Their genotypes may not reflect those discussed in the strain description. Please inquire for possible genotypes and see additional details below.
Standard Supply
Cryopreserved. Ready for recovery. Please refer to pricing and supply notes on the strain data sheet for further information.
Supply Notes
- Cryorecovery - Standard.
Progeny testing is not required.
The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
Mice recovered can be used to establish a dedicated colony to contractually supply you mice according to your requirements. Price by quotation.
