Mice homozygous for both the Tnfrsf1atm1Imx and Tnfrsf1btm1Imx targeted mutations (p55 and p75 deficient) are viable and fertile. Double homozygous mutant mice fail to bind TNF. Thymus, spleen, and other lymphoid tissue are normal, indicating that TNF is not required for normal development of these organs.Development
The double mutant mice lacking both Tnfrsf1a (p55) and Tnfrsf1b (p75) were generated by intercrossing the singly deficient mice. According to the donating investigator, the Tnfrsf1b mutation (129S7/SvEvBrd background) was moved onto the Tnfrsf1a background (C57BL/6) by successive backcrosses to the latter (to N4). For the Tnfrsf1a mutation, a PGK-neomycin resistance cassette replaced 4kb of sequence, including exons 2 to 5 (amino acids 30 - 184). For the Tnfrsf1b mutation, a PGK-neomycin resistance cassette replaced 1kb of sequence encoding amino acids 3 - 26.
