| General Note | Three transgenic lines were established. This line and the line originally called TgN(HuZP3)2NIH each was estimated by Southern blot densitometry to carry 2 copies, and the line originally called TgN(HuZP3)1NIH to carry 4-6 copies of the transgene. Authors state that null-allele rescue results using the former two strains were identical. | ||
| Molecular Note | This transgene comprises 6 kb of 5' flanking DNA from the mouse zona pellucida 3 gene ligated to the 5' half of the human zona pellucida 3 gene, including exons and introns 1-4, followed by cDNA encoded by exons 5-8 of the human gene and the bovine growth hormone polyadenylation signal. This transgenic line was estimated by Southern blot densitometry to carry two copies of the transgene. Transgene expression was shown by RNase protection analysis of multiple tissues to be ovary-specific. In situ hybridization localized expression to the cytoplasm of growing oocytes, at a level similar to that of the co-expressed endogenous mouse gene. Species-specific antibodies detect lower levels of human and mouse ZP3 in zonae pellucidae of mature transgenic oocytes than in human or wild-type mouse zonae, respectively; the total (human + mouse) approximating that in normal mouse zonae. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. The total number of animals provided, their gender and genotype will vary. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 13 and 16 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
