Former Names FVB/NJ-TgN(XGFAPLacZ)3Mes (Changed: 15-DEC-04 ) Type Mutant Strain; Transgenic; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Species laboratory mouse Generation +N1 Donating Investigator IMR Colony, The Jackson Laboratory Appearance
albino
Related Genotype: Tyrc/TyrcImportant Note
This strain is homozygous for the retinal degeneration allele Pde6brd1.Description
Mice carrying this transgene express beta galactosidase in the nuclei of astrocytes. The donating investigator reports that more recent studies on this particular line indicate low level expression of the lacZ reporter gene in many neuronal populations as well. The transgene integrated on the X chromosome and therefore undergoes X-inactivation. This strain may be useful for cell culture and transplantation studies.Development
This strain was developed in the laboratory of Dr. Albee Messing at the University of Wisconsin School of Veterinary Medicine. The construct contains base pairs -2163 to +47 relative to the transcriptional start site of human GFAP, and has the normal ATG start site at +15 converted to TTG by site-directed mutagenesis so that protein synthesis initiates within the lacZ coding region. The construct results in targeting of the enzyme to the nucleus. The transgene was injected into B6SJL/F2 mice and integrated onto the X chromosome.

| General Note | Mice carrying this transgene express beta galactosidase in the nuclei of astrocytes. Transgenic mice exhibit upregulation of GFAP gene activity similar to that which follows injury to the brain. The transgene inserted on the X Chromosome and appears to undergo the usual random inactivation that achieves gene dosage compensation in females. The brains of hemizygous females stain uniformly rather than displaying mosaic patches, suggesting that astrocytes intermingle following their formation. | ||
| Molecular Note | The transgene contains a bacterial lacZ gene linked to a 2.2 kilobase 5'-flanking sequence derived from the human glial fibrillary acidic protein gene. The human GFAP promoter sequence has been shownto direct astrocyte-specific transcription in culturedcells. [MGI Ref ID J:92866] | ||
