Type Mutant Stock; Transgenic; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse Generation N2 Donating Investigator Lilia Stepanova, St. Jude Children's Research Hospital Description
In wildtype mice Cdc37 is expressed primarily in proliferative tissues. These transgenic mice express Cdc37 under the direction of an MMTV promoter. As a result, levels of transgene mRNA are significantly higher than levels of endogenous Cdc37 in specific tissues (lacrimal, mammary, salivary glands, uterus and testis). Mice are born and develop normally. By 18 months of age females exhibit proliferative disorders including mammary tumors and lymphomas. By 22 months of age 100% of transgenic females develop mammary or lymphoid tumors. Histopathological examination indicates that mammary tumors are adenocarcinomas and adenosquamous carcinomas. Males expressing Cdc37 in mammary tissue develop a system of mammary ducts resembling that of virgin females, but do not develop tumors. Crosses with other MMTV-oncogene expressing mice indicate that Cdc37 can cooperate with myc in transformation of multiple tissues, and with Cyclin D1 in transformation of the mammary epithelium.Development
A plasmid containing a MMTV promoter, rabbit beta globin splice sequence, mouse Cdc37 open reading frame, and bovine polyadenylation signal was injected into male pronuclei of B6D2F1 mouse embryos. Founder animals were mated with ICR mice.
