Type JAX® GEMM® Strain - Mutant Strain Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Transgenic Species laboratory mouse Donating Investigator Joshy Jacob, Emory University School of Medicine Generation N3+1F4pN1 Strain Description
This strain is one component of a Cre-recombinase mediated mouse model system that allows irreversible labeling of antigen-stimulated T cells with surface expressed ALPP (placental alkaline phosphatase). Cre recombinase is expressed under control of the human GZMB (granzyme B) promoter which directs specific expression in activated T cells. When this strain is bred with a Cre-recombinase expressing strain the loxP-flanked STOP fragment is excised. This strain represents an effective tool for generating tissue specific-targeted mutants.Strain Development
A targeting construct containing the human granzyme B promoter, the SV40 nuclear localization signal sequence, Cre recombinase, and the human growth hormone polyadenylation sequence was injected into the pronuclei of fertilized FVB/N eggs. Founder mice were obtained and bred.

| Site of Expression | activated T cells | ||
| Expressed Gene | cre, cre recombinase, bacteriophage P1 | ||
| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||
| Promoter | GZMB, granzyme B (granzyme 2, cytotoxic T-lymphocyte-associated serine esterase 1), human | ||
| Molecular Note | This transgene expresses Cre recombinase under the control of a human granzyme B (GZMB) promoter, active specifically in activated T cells. [MGI Ref ID J:67937] | ||
Generic Cre
| Breeding & Husbandry | This mutation was generated on a FVB/N background. Founder animals were backcrossed to C57BL/6 (N3 5/2000). |

Cryorecovery - Standard. The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery. Cryorecovery to establish a Dedicated Supply for greater quantities of mice. |
