Former Names NOD/Lt-Tg(TcraAI4)1Dvs/+ Tg(TcrbAI4)1Dvs/+ (Changed: 23-FEB-07 ) Type Coisogenic; Mutant Strain; Transgenic; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse H2 Haplotype g7 Generation F1 (18-DEC-06) Appearance
albino, pink eyed
Related Genotype: A/A Tyrc/TyrcDevelopment
This strain is an obligate double hemizygous F1 made by mating NOD/Lt-Tg(TCRaAI4)1Dvs (Stock #004334) and NOD/Lt-Tg(TCRbAI4)1Dvs (Stock #004335). This strain must be generated in this manner because mice carrying both the alpha and beta transgenes rapidly develop diabetes. (Graser et al., 2000.)

NOD-Tg(TcraAI4)1Dvs Tg(TcrbAI4)1Dvs
- immune system phenotype
- abnormal CD4-positive T cell morphology (MGI Ref ID J:93553)
- T cells expressing the transgenes comprise ~35% of the total peripheral blood lymphocyte population, a significant fraction of these T cells resides in the CD4 lineage rather than in the expected CD8 lineage
- abnormal CD8-positive T cell morphology (MGI Ref ID J:94192)
- only transgene expressing CD8+ T cells from primary splenocytes expand when cultured in presence of islets from NOD-scid donors
- abnormal cytotoxic T cell physiology (MGI Ref ID J:94192)
- transgenic CTLs generated in culture on islets exhibit cytotoxicity against a NOD-derived beta cell adenoma cell line and this is augmented when adenoma cells are pretreated with IFNG
- increased susceptibility to autoimmune diabetes (MGI Ref ID J:94192)
- female NOD mice carrying both transgenes develop IDDM at a greatly increased rate compared to nontransgenic controls with disease appearing as early as 3 weeks of age
- hematopoietic system phenotype
- abnormal CD4-positive T cell morphology (MGI Ref ID J:93553)
- T cells expressing the transgenes comprise ~35% of the total peripheral blood lymphocyte population, a significant fraction of these T cells resides in the CD4 lineage rather than in the expected CD8 lineage
- abnormal CD8-positive T cell morphology (MGI Ref ID J:94192)
