Former Names B6;129S-Mtap7tm1Sor/J (Changed: 10-JAN-05 ) Type Gene Trap; Mutant Stock; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse Generation N3 Donating Investigator Philippe Soriano, Fred Hutchinson Cancer Research Center Description
At birth, mice homozygous for the gene-trapped Mtap7 allele are viable, normal in size and do not display any gross physical or behavioral abnormalities. Although trace amounts of a presumably nonfunctional transcript can be detected in testis tissue, no protein product is immunodetectable. Male homozygotes are sterile. Expression of the reporter gene (B-galactosidase from the Bgeo fusion gene) employed by the gene trap vector indicates that the Mtap7 promoter directs expression in various tissues with highest levels seen in the seminiferous tubules. During the first wave of spermatogenesis at 5 weeks of age, deformed spermatids can be observed. Abnormalities are attributed to aberrant microtubule organization. Microtubule aberrations are also observed in Sertoli cells. Gradual loss of germ cells occurs. At three months of age, the testes of homozygous mutants are less than one-third the size of those of heterozygous littermates.Development
A retroviral genetrap vector (ROSABgeo*) containing a Bgal/neo fusion gene (Bgeo) was used to infect 129S4/SvJaeSor-derived AK7 embryonic stem (ES) cells. ES cells expressing neo (G418 resistant) were selected. Cells were subjected to an additional round of screening for retinoic acid (RA) responsiveness. RA responsive cells were injected into C57BL/6 blastocysts and the resulting chimeric mice were crossed to C57BL/6 animals. The vector was found to have inserted into the 5' portion of the Mtap7 gene.
