Mice that are homozygous for this targeted allele appear weak, fail to feed and die within a few hours of birth from unspecified causes. Embryos exhibit a slightly foreshortened head and abnormal lens development. No Maf gene transcript is detected. Posterior lens cells from homozygotes fail to elongate. These cells display inappropriately high levels of DNA synthesis and express alpha A crystallin and all of the beta crystallin genes at dramatically reduced levels. Heterozygotes are phenotypically normal. The presence of lacZ in the targeted allele makes it possible to observe Maf expression characteristics.Development
A targeting vector containing neomycin resistance, lacZ and herpes simplex virus thymidine kinase genes was used to disrupt the single Maf exon. The targeting construct was designed to remove the carboxy-terminal 351 amino acids and 3kb of 3' untranslated sequence. The lacZ gene was fused to an amino-terminal portion of the Maf gene encoding 19 amino acids. The construct was electroporated into 129X1/SvJ x 129S1/Sv-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric animals were backcrossed to C57BL/6J mice.

We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. The total number of animals provided, their gender and genotype will vary. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 13 and 16 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
