Type Congenic; Mutant Strain; Targeted Mutation; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Additional information on Congenic nomenclature. Species laboratory mouse Generation N2 Donating Investigator Greg Barsh, Stanford University Medical Center Description
Mice that are homozygous for this targeted allele appear weak, fail to feed and die within a few hours of birth from unspecified causes. Embryos exhibit a slightly foreshortened head and abnormal lens development. No Maf gene transcript is detected. Posterior lens cells from homozygotes fail to elongate. These cells display inappropriately high levels of DNA synthesis and express alpha A crystallin and all of the beta crystallin genes at dramatically reduced levels. Heterozygotes are phenotypically normal. The presence of lacZ in the targeted allele makes it possible to observe Maf expression characteristics.Development
A targeting vector containing neomycin resistance, lacZ and herpes simplex virus thymidine kinase genes was used to disrupt the single Maf exon. The targeting construct was designed to remove the carboxy-terminal 351 amino acids and 3kb of 3' untranslated sequence. The lacZ gene was fused to an amino-terminal portion of the Maf gene encoding 19 amino acids. The construct was electroporated into 129X1/SvJ x 129S1/Sv-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric animals were backcrossed to C57BL/6J mice.

The following phenotype information may relate to a genetic background differing from this JAX® Mice strain.
Maftm1Gsb/Maftm1Gsb
involves: 129S1/Sv * 129X1/SvJ * C57BL/6J
- lethality-prenatal/perinatal
- neonatal lethality (MGI Ref ID J:56583)
- mice die within hours of birth
- vision/eye phenotype
- abnormal cornea morphology (MGI Ref ID J:56583)
- in older mice, corneas are thickened and exhibit increased cell proliferation and irregularity at the endothelial surface
- abnormal lens development (MGI Ref ID J:56583)
- by E12.5, fiber cell differentiation and elongation ceases
- early maturation of primary and secondary lens fiber cells and the ability of these cells to remain in G0 is disrupted
- at E15.5, the boundary between proliferating and non-proliferating cells is shifted anteriorly
- abnormal lens vesicle development (MGI Ref ID J:56583)
- by E13.5, the lens vesicle is hollow whereas it is filled in wild-type mice
- by E15.5, epithelial cells are confined to the anterior third rather than the anterior half of the lens vesicle
- after E16.5, the lens vesicle is deformed
- small lens (MGI Ref ID J:56583)
- at E17.5, lens size is reduced by one half or less of wild-type
- craniofacial phenotype
- shortened head (MGI Ref ID J:56583)
- mice exhibit a fore-shortened head at birth
- behavior/neurological phenotype
- decreased eating behavior (MGI Ref ID J:56583)
- mice do not eat at birth
- weakness (MGI Ref ID J:56583)
- at birth
