Strain Development
This strain is an obligate double hemizygous F1 which is homozygous for Ragtm1Mom. It is made by mating NOD.Cg-Rag1tm1Mom Tg(TcraAI4)1Dvs (Stock #004232) with NOD.Cg-Rag1tm1Mom Tg(TcrbAI4)1Dvs (Stock #004233). This strain must be generated in this manner because mice carrying both the alpha and beta transgenes rapidly develop diabetes.

  • immune system phenotype
  • increased susceptibility to autoimmune diabetes (MGI Ref ID J:94192)
    • females have a greatly increased rate of diabetes development compared to non-transgenic NOD controls (100% incidence by 6 weeks of age)

Gene Symbol and Name Rag1, recombination activating gene 1
Chromosome 2
Gene Common Name(s) MGC43321; RNF74; Rag-1;
Molecular Note A 1356 bp genomic fragment of the Rag1 gene, encoding the nuclear localization signal and the zinc-finger motif, was replaced by a neomycin cassette. A mutant transcript expressed from this allele was detected by Northern blot in bone marrow derived cell lines from homozygous mice. [MGI Ref ID J:1934] [MGI Ref ID J:96036]

Allele Symbol Tg(TcraAI4)1Dvs
Allele Name transgene insertion 1, David V Serreze
Common Name(s) AI4aTg;
Mutation Made By David Serreze,   The Jackson Laboratory
Strain of OriginNOD/ShiLt
Expressed Gene Tcra, T-cell receptor alpha chain, mouse, laboratory
General Note In conjunction with Rag1tm1Mom
    transgenic mice carrying both the alpha and beta AI4 TCR transgenes exhibit an accelerated development of insulin-dependent diabetes mellitus (IDDM).
    homozygous mice are viable and fertile
Molecular Note The rearranged Tcra transgene expressed in these mice is derived from the pancreatic beta cell-reactive CD8 T lymphocyte clone AI4. The transgene contains a Tcra-V8/Tcra-J cDNA segment. [MGI Ref ID J:93553]

Allele Symbol Tg(TcrbAI4)1Dvs
Allele Name transgene insertion 1, David V Serreze
Common Name(s) AI4bTg;
Mutation Made By David Serreze,   The Jackson Laboratory
Strain of OriginNOD/ShiLt
Expressed Gene Tcrb, T-cell receptor beta chain, mouse, laboratory
General Note In conjunction with Rag1tm1Mom
    transgenic mice carrying both the alpha and beta AI4 TCR transgenes exhibit an accelerated development of insulin-dependent diabetes mellitus (IDDM).
    homozygous mice are viable and fertile
Molecular Note The rearranged Tcrb transgene expressed in these mice is derived from the pancreatic beta cell-reactive CD8 T lymphocyte clone AI4. The transgenic construct contains a Tcrb-V2/Tcrb-D/Tcrb-J cDNA segment. [MGI Ref ID J:93553]

The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
