These transgenic mice express Enhanced Green Fluorescent Protein (EGFP) under the direction of the mouse Purkinje cell protein 2 promoter. Mice hemizygous for the transgene are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Transgenic mice display distinct expression of EGFP in dendrites, axons and soma of Purkinje cells, allowing identification, isolation and purification of Purkinje cells by FACS. EGFP expression mimics endogenous Purkinje cell protein 2 expression pattern. Fluorescence is detectable at embryonic day 17 in Purkinje cells and also occurs in retinal bipolar cells. Low levels of fluorescence are seen in olfactory periglomerular cells, interpeduncular nucleus and superior colliculus neurons. This mutant mouse strain represents a model that may be useful for purification and in vivo tracking of Purkinje cells.Strain Development
A transgenic construct containing an Enhanced Green Fluorescent Protein (EGFP, Clontech) gene under the control of a mouse Purkinje cell protein 2 promoter was injected into fertilized FVB/N mouse eggs. The donating investigator intercrossed founder lines 146 and 244 prior to the strain's arrival at The Jackson Laboratory. During the development of the production stock at The Jackson Laboratory, the founder insert with the larger number of transgene copies was retained as two more backcrosses to C57BL/6J were performed. It is unknown if the retained insert originated from founder line 146 or 244.

The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP).
