Tg(Ins2-tTA)1Doi encodes the tetracycline regulatable transactivator (tTA) regulated by the rat insulin promoter (Ins2, commonly designated RIP), and is expressed in the pancreatic beta cells. Mice hemizygous for this transgenic insert are viable, fertile, normal in size, display normal NOD diabetes onset, but do not display any other gross physical or behavioral abnormalities. When transgenic mice are mated to a second transgenic strain carrying the target gene regulated by the tetO responsive elements, expression of the target gene is turned off by the tetracycline analog, doxycycline (dox). Dox can be administered orally in the food or water. This strain provides a Tet-Off tool that permits the inducible expression of genes in the pancreatic beta cells during various stages of diabetes development.Development

| The tetracycline-resistance gene (TetR), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). TetR was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). The tetracycline-inhibitable transcription factor is a component of a bigenic system that allows doxycycline (a tetracycline analog) regulatable expression of genes that are under the direction of the tetracycline responsive promoter (TetOp)promoter. | |||

Breeding & Husbandry When held in a live colony, this strain is maintained Tg/? X Tg/? as we are unable to determine hemizygotes from homozygotes, A footpad injection of Complete Freund s Adjuvant (CFA) administered once at weaning will delay diabetes onset, thus extending the lifespan of breeders. Use of Complete Freund s Adjuvant in NOD mice can be found in Current Protocols in Immunologypage 15.9.19, Reproduction.
