Strain Description
Mice carrying this "-408SP10-gfp" transgene express Enhanced Green Fluorescent Protein (EGFP) under the direction of the -408 to +28 bp mouse SP-10 (Acrv1) promoter region that contains the necessary transcription regulation signals responsible for both temporal and testis-specific gene expression in transgenic mice. In hemizygous mice, EGFP fluorescence is detected in postmeiotic round spermatid cells of testis beginning from post natal day 21 through adult life. Transgene expression is detected in testis by by northern hybridization as well as RT-PCR analysis. In situ hybridization examination of testis from transgenic mice reveals EGFP is expressed in a pattern mimicking endogenous Acrv1 spatial and temporal expression. This strain may be useful in studies related to spermatogenesis.

Strain Development
A transgenic construct containing the -408 to +28 bp promoter fragment from the mouse SP-10 (Acrv1) gene, an Enhanced Green Fluorescent Protein (EGFP; Clontech) open reading frame, and SV40 polyadenylation site sequence was injected into fertilized B6CBAF2 mouse eggs. Transgenic founder animals (line 2727) were backcrossed for 3 generations on the C57BL/6 background.

Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source.

General Note Two of three transgenic founder mice expressed EGFP in testis only; the third failed to express it in any tissue. No distinction was made between the two expressing lines.
Molecular Note This transgene contains a cDNA encoding enhanced green fluorescent protein, with an SV40 polyadenylation signal, downstream of a promoter fragment from the mouse acrosomal vesicle protein 1 gene encompassing nucleotides -408 through +28 (relative to the transcription start site). 21-day-old, but not 18-day-old male transgenic mice exhibit EGFP fluorescence specifically in seminiferous tubules, initially in postmeiotic, round spermatids. Experienced technicians at The Jackson Laboratory have recovered frozen embryos of this strain successfully. We will provide you enough embryos to perform two embryo transfers. Cryorecovery - Standard.
The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery.

The Jackson Laboratory's Genotype Promise

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
