These transgenic mice express Cre recombinase under the control of the mouse Fabp4, fatty acid binding protein 4, adipocyte, promoter. Cre recombinase expression is detected in brown and white gonadal and subcutaneous adipose tissue. No expression is detected in skeletal muscle. When crossed with a strain containing a loxP site flanked sequence of interest, Cre-mediated recombination results in deletion of the targeted gene specifically in adipose tissue. Mice that are homozygous for the targeted mutation are viable. This strain represents an effective tool for generating tissue-specific targeted mutants.Strain Development
A transgenic construct containing cre coding sequence under the control of the 5.4 kb Fabp4, fatty acid binding protein 4, adipocyte, promoter was introduced into unknown outbred donor eggs. The resulting male transgenic mice were crossed to B6.129-Ppargtm2Rev/J (Stock No. 004584) mice and subsequently crossed to C57BL/6J mice for 9 generations in the laboratory of Dr. Yaacov Barak at The Jackson Laboratory. The Ppargtm2Rev/J allele was bred out of the strain during the backcrossing to C57BL/6J.

| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||

| Molecular Note | Sequence encoding cre recombinase was adjoined to the Fabp4 promoter, which is active in white and brown adipose tissue. Northern blot analysis and a functional assay showed the transgene to be abundantly expressed in brown and white adipose tissue. | Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged.
