This NOD/LtJWehi congenic strain is carrying Chr 1alleles derived from BALB/cWehi including Ptprcb. Diabetes onset and incidence is similar between NOD.C-(Ptprc-D1Mit262)/WehiJ congenic mice (6/8 by 300 days of age) and NOD/LtJWehi (17/24 by 300 days of age). NOD.C-(Ptprc-D1Mit262)/WehiJ bone marrow transplanted into irradiated NOD/LtJWehi recipients yielded incomplete replacement of donor leukocytes at 16 weeks post transfer versus nearly complete replacement of donor leucocytes in C57BL/6Wehi and B6.SJL/Wehi irradiated mice (Steptoe, et al., 2004, Journal of AutoImmunity, Vol 22, p131-138). This strain provides a unique cell surface marker that can be used to track lineage and phenotype of all hematopoietic donor derived or transferred cells making it useful in studies monitoring diabetes progression.Development
Ptprcb a Chr 1 genomic segment from BALB/cWehi was transferred to NOD/LtJWehi (Ptprca). An (NODxBALB/c)F1 male was backcrossed to NOD/LtJWehi followed by 10 backcross cycles to NOD/LtJWehi with selection for BALB/c markers on Chr. 1 prior to inbreeding. FACS analysis indicates Ptprc (74cM) is BALB/c in origin (Ptprcb). Microsatellite analysis indicates D1Mit262 (69.9cM) is BALB/c in origin while D1Mit494 (68.9cM) and D1Mit102 ((75.4cM) are of NOD/LtJWehi origin. Additionally, microsatellite analysis confirms all known Idd alleles (1-20) are NOD in origin (Steptoe, et al., 2004, Journal of AutoImmunity, Vol 22, p131-138). Breeding & Husbandry A footpad injection of Complete Freund s Adjuvant (CFA) administered once at weaning will delay diabetes onset, thus extending the lifespan of breeders. Use of Complete Freund s Adjuvant in NOD mice can be found in Current Protocols in Immunology page 15.9.19, Reproduction.
