Type JAX® GEMM® Strain - Congenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Transgenic Species laboratory mouse Donating Investigator Joshua Sanes, Harvard University Generation N7p (05-FEB-06) Strain Description
These transgenic mice express a fusion gene consisting of synaptophysin and Yellow Fluorescent Protein (EYFP) under the direction of the mouse thymus cell antigen 1, theta promoter. Expression of EYFP is detected in vestibular and pontine neurons by birth to 2 days of age. Fluorescent signal is punctuate in the mossy fibers of cerebellar granule layer. The donating investigator reports high levels of fluorescence is detected in synaptic vesicles of neuromuscular junctions. Mice that are hemizygous for the targeted mutation are viable, normal in size and do not display any gross physical or behavioral abnormalities. This mutant mouse strain may be useful in studies of synapse formation at nerve terminals.Strain Development
A transgenic construct containing a fusion gene consisting of synaptophysin and Yellow Fluorescent Protein (EYFP) under the direction of the mouse thymus cell antigen 1, theta promoter was injected into fertilized B6CBAF1 mouse eggs. The resulting transgenic animals of founder line 10 were backcrossed to C57BL/6 mice for 6 generations.

| Site of Expression | Vestibular and pontine neurons by birth to 2 days of age; punctuate in the mossy fibers of cerebellar granule layer; high levels in synaptic vesicles of neuromuscular junctions. | ||
| Expressed Gene | YFP, Yellow Fluorescent Protein, jellyfish | ||
| Yellow Fluorescent Protein (YFP) is a derivative of Green Fluorescent Protein (GFP), a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance fluorescence intensity and shift the wavelength emitted when excited. When YFP is utilized in a transgenic construct, tissue expressing sufficient amounts of YFP will fluoresce yellowish-green when exposed to a 513 nm light source. | |||
| Expressed Gene | Syp, synaptophysin, mouse, laboratory | ||
| Promoter | Thy1, thymus cell antigen 1, theta, mouse, laboratory | ||
| General Note | From founder lines in which the mossy fibers expressed YFP, line 10 was selected because the pontine and vestibular neurons of these mice begin to express YFP between birth and postnatal day 10. | ||
| Molecular Note | Mice with this transgene express a fusion protein composed of synaptophysin and enhanced yellow fluorescent protein under control of the neuron-specific regulatory elements of the mouse thymus cell antigen 1, theta (Thy1) gene. [J:91948] | ||

Cryorecovery - Standard. The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery. Cryorecovery to establish a Dedicated Supply for greater quantities of mice. |
