This transgenic strain expresses a variant of the Saccharomyces cerevisiae FLP1 recombinase gene under the direction of the human ACTB promoter. The FLPe recombinase variant exhibits enhanced thermostability with recombination activity being four-fold and ten-fold that of wildtype FLP at 37oC and 40oC, respectively. Mice that are hemizygous for the transgenic allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Recombinase activity is detected in a wide variety of tissues (spinal cord, heart, gonad, adrenal) as early as embryonic day 10.5. This deleter strain is a suitable alternative to, and complement with the Cre-loxP system for in vivo genetic engineering.In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Development
A transgenic construct containing a FLP1 recombinase variant controlled by the human ACTB promoter (3 kb flanking sequence, 78-bp 5' untranslated region, 832-bp of exon 1) was injected into fertilized eggs from B6SJLF2 mice. Founder animals were obtained and crossed with C57BL/6J mice.

| FLP1 recombinase is an enzyme derived from Saccharomyces cerevisiae that specifically recognizes a 34 base pair FLP1 recombinase target (FRT) sequence. FLP1 has been shown to effectively mediate the excision of DNA located between similarly oriented FRT sites. After the excision event, DNA ends recombine leaving a single FRT site in place of the intervening sequence. DNA located between oppositely orientated FRT sites is inverted rather than deleted. | |||
