Strain Description
Mice homozygous for both the targeted mutation and the transgene are viable and fertile. The transgene beta-galactosidase is well expressed, reflects endogenous Tek/Tie2 activity, and is specific to the vascular endothelium. Mice homozygous for the Rag1 mutation are immunodeficient as they lack mature B and T lymphocytes. This double mutant mouse may be useful in studies of cancer, tumor-related angiogenesis, and as a recipient of xenografted tumors.

Strain Development
The transgene consists of a beta-galactosidase reporter gene under the control of a modified murine endothelial-specific receptor tyrosine kinase (Tek/Tie2) promoter. Targeted mutation of recombination activating gene-1 (Rag1) resulted in a 1356 base pair deletion in the 5' end of the endogenous coding sequence and PGK-Neo cassette insertion. Double mutant mice were generated by crossing homozygous female B6.129S7-Rag1tm1Mom/J (see Stock No. 002216) with hemizygous male FVB/N-Tg(TIE2-lacZ)182Sato/J (see Stock No. 002856) mice. Female progeny were then mated to males homozygous for the Rag1 mutation to fix Rag1-deficiency. Subsequent sibling intercross resulted in presumed homozygosity of the transgene on chromosome X (via test crosses to wildtype). Other transgene insertion points on autosomes are likely.

Colony Maintenance

Breeding & HusbandryWhen maintaining a live colony, these mice are bred as homozygous for both the targeted mutation and the transgene. Specific pathogen free conditions are recommended as these mice are immunodeficient (lack mature B and T lymphocytes). Donating investigator reports occasional abnormalities at their facility; mice can agressively barber their cagemates, have tooth malocclusion at weaning, and a few develop 'crystal pneumonitis'.

This strain is also available as cryopreserved embryos from our Repository. Experienced technicians at The Jackson Laboratory have recovered frozen embryos of this strain successfully. We will provide you enough embryos to perform two embryo transfers. Cryorecovery - Standard.
The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery.
