Tg(Ins2-rtTA)2Ach encodes the reverse tetracycline regulatable transactivator (rtTA) regulated by the rat insulin promoter (Ins2, commonly designated RIP), and is expressed in the pancreatic beta cells. Hemizygous transgenic mice are viable, fertile, normal in size, display normal NOD diabetes onset, but do not display any other gross physical or behavioral abnormalities. When NOD/Lt-Tg(Ins2-rtTA)2Ach/AchJ mice are mated to a second transgenic strain carrying a target gene regulated by the tetO responsive elements, expression of the target gene is turned on by the tetracycline analog, doxycycline (dox). Dox can be administered orally in the food or water. This strain provides a Tet-On tool that permits the inducible expression of genes in the pancreatic beta cells during various stages of diabetes development (As reported by donating investigator, data unpublished).This transgenic strain is likely to have characteristics similar to Stock No. 004602. Note that the genetic background this strain is comprised entirely of NOD/ShtLt whereas Stock No. 004602 background is largely NOD/ShiLt but may have contributions from the C57BL/6J and SJL inbred strains.
Development
Alexander Chervonsky, et al., injected the Ins2-rtTA transgenic construct directly into NOD/ShiLt oocytes. The resulting transgenic line has been maintained by continual matings to NOD/ShiLt. In 2005, The T1DR received this NOD/ShiLt transgenic which is likely to have characteristics similar to Stock No. 004602. Note that the genetic background of this strain is comprised entirely of NOD/ShiLt whereas Stock No. 004602 background is largely NOD/ShiLt but may have contributions from the C57BL/6J inbred strain.

| The tetracycline repressor gene (Tetr), arose from chemically mutated Escherichia coli genome which was screened for tetracycline dependence (Gossen and Bujard, 1992). One mutant with a four amino acid residue change (rTetR) exhibited dependence on tetracycline for induction of the targeted gene and was used in the rtTA construct (Gossen et al, 1995). rTetr was fused at the C-terminus with the viral co-activator, virion protein 16 of the herpes simplex virus (VP-16). | |||

IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
