Description
Hemizygous offspring from wild type mothers and transgenic fathers are viable, fertile, and normal in size. The imprinted methylation pattern on this transgene is dependent on maternal or paternal transgene inheritance, and is faithful to normal patterns with the following exceptions; defective methylation of Tssc4 and improper imprinting of Tssc4 and Ascl2. Fetal growth restriction is observed following maternal, but not paternal, transmission of the transgene. This phenotype is a characteristic of maternal duplication of chromosome 11p15.5. In addition, this mouse may also be useful in studying Beckwith-Wiedemann syndrome, imprinting/methylation mechanisms, and differentially methylated regions.

Development
The 800 kb yeast artificial chromosome (YAC) W408A5 containing C57BL/6 mouse genomic sequence from distal chromosome 7 (extending from the Cars to the Th gene and including imprinting center 2 (IC2), Cdkn1c (P57), and Kcnq1) was modified to by addition of two copies of a neomycin-selection cassette. This construct was introduced into 129/Sv-derived embryonic stem (ES) cells by yeast spheroplast fusion. Correctly targeted ES cells containing a full length and single copy YAC were injected in C57BL/6 blastocysts which were then implanted into pseudopregnant foster mothers. Chimeric founder line L1952 males were backcrossed 5 generations to SD7 female before arrival at The Jackson Laboratory. SD7 mice (M. spretus distal chromosome 7 on C57BL/6 M. Domesticus background) were used to provide a source of polymorphisms to distinguish expression and methylation of all the genes present on the transgene.

Molecular Note An 800kb yeast artificial chromosome (YAC) designated W408A5, which contained C57BL/6 distal chromosome 7 sequence from the Cars to Th gene, and including imprinting center(IC2 which contains Cdkn1c (P57) and Kcnq1) and is syntenic to human chromosome 11p15.5, was modified by addition of 2 copies of the Neo-selection cassette. The construct was introduced into 129/Sv ES cells by yeast spheroplast fusion.After maternal transmission of the transgene, most maternally expressed genes of the IC2 region (Phdla2, Slc22a18, Cdkn1c and Kcnq1) show 30-60% increased expression in the placenta relative to wild-type and equivalent or slightly reduced expression after paternal transmission. With paternal transmission, paternally expressed gene Kcnq1ot1 shows 2-fold increased expression over wild-type or after maternal transmission. Ascl2 expression (maternally expressed) is increased 2-fold after maternal or paternal transmission, while Tssc4 expression is increased 2-fold after maternal and by ~33% after paternal transmission. Imprinting of these 2 genes is not reproduced by the transgene. "MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCTS" means biological materials supplied by JACKSON, and their derivatives. "RECIPIENT" means each recipient of MICE, PRODUCTS, or services provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE or PRODUCTS from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON's prior written authorization.
