Type JAX® GEMM® Strain - Congenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Transgenic Mating System Hemizygote x +/+ sibling (Female x Male) Species laboratory mouse Donating Investigator Brian Popko, University of Chicago Generation N10+1F4 (08-JAN-08) Strain Description
These transgenic mice have a tamoxifen inducible Cre-mediated recombination system driven by the mouse Plp1, proteolipid protein (myelin) 1 promoter. The transgene insert contains a fusion product involving Cre recombinase and a mutant form of the mouse estrogen receptor ligand binding domain. The mutant mouse estrogen receptor does not bind natural ligand at physiological concentrations but will bind the synthetic ligand, 4-hydroxytamoxifen. Restricted to the cytoplasm, the Cre/Esr1 protein can only gain access to the nuclear compartment after exposure to tamoxifen. When crossed with a strain containing a loxP site flanked sequence of interest, the offspring are useful for generating tamoxifen-induced, Cre-mediated targeted deletions. Tamoxifen administration allows for ablation of predetermined genes in oligodendrocytes and Schwann cells, and will also induce Cre recombination in developing embryos of treated mothers and in cultured cells derived from transgenic mice. Mice hemizygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Immunohistochemical analysis of brain, optic and sciatic nerve tissue reveals that the cre/Esr1 expression mimics the endogenous promoter expression pattern.Strain Development
A transgenic construct containing a 2.4 kb sequence of mouse proteolipid protein (myelin) 1 promoter, sequence encoding cre/Esr1 and SV40 polyadenylation site sequence was injected into B6D2F1 donor eggs. Founder animals were bred to C57BL/6J mice and then backcrossed to the same for 10 generations.

| Expressed Gene | cre, cre recombinase, bacteriophage P1 | ||
| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||
| Promoter | Plp1, proteolipid protein (myelin) 1, mouse, laboratory | ||
| Molecular Note | The transgenic construct encodes a tamoxifen inducible fusion protein consisting of cre recombinase and a mutated human estrogen receptor ligand binding domain cloned next to a Plp promoter. The 2.5 kb Plp regulatory region includes 5' flanking DNA, exon1, and intron 1. Transgene expression was confirmed by RT-PCR and Western blot analysis. The mouse Plp promoter was used to drive transgene expression in myelinating cells. [MGI Ref ID J:81627] | ||

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
