Mice homozygous for this Actb-DsRed.T3 transgene are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express the red fluorescent protein variant DsRed.MST under the control of the chicken beta actin promoter coupled with the cytomegalovirus (CMV) immediate early enhancer. All tissues of homozygotes fluoresce red. Mice hemizygous for the transgene express red fluorescent protein less intensely than homozygotes. Expression is observed throughout all embryonic and adult stages and very high expression is found in pancreas, skeletal muscle, heart and seminal vesicle.In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description as published results become available.
Development
This strain is a derivative of Tg(ACTB-bgeo/DsRed.MST). A transgenic construct containing a Red Fluorescent Protein gene (DsRed.MST) under the control of the a chicken beta actin promoter coupled with the cytomegalovirus (CMV) immediate early enhancer upstream of a loxP site flanked beta-geo gene and polyadenylation sites, was introduced into 129S6B6F1 derived G4 embryonic stem (ES) cells. Then the expression of lacZ was replaced with red fluorescent protein following in vitro Cre recombination. ES cells containing the (ACTB-DsRed.MST) construct were aggregated with ICR outbred tetraploid embryos to generate chimeric mice. The resulting chimeric male animal was bred with an ICR outbred female. Progeny from this cross that were hemizygous for the transgene were bred to ICR outbred mice. The mice were then backcrossed to C57BL/6J for 5 generations. Of note, this "DsRed.MST" protein is erroneously designated "DsRed.T3" in the original publication.
