Type Congenic; Mutant Strain; Transgenic; Additional information on Genetically Engineered Mutant Mice. Mating System +/+ sibling x Hemizygote (Female x Male) Species laboratory mouse Generation ?+N5+F0 (24-JUL-07) Donating Investigator IMR Colony, The Jackson Laboratory Description
Mice homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) under the control of the chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer. Nucleosomes and chromatin in all cells fluoresce. Fluorescence is detectable during all phases of mitosis. The donating investigator reports occasional silencing of the transgene when transmitted through the female germline. This mutant mouse strain may be useful for in vivo subcellular high-resolution and 3-dimensional studies of dividing cells. In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Development
A transgenic construct containing an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) was placed under the control of a chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer promoter. This construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-derived R1 embryonic stem (ES) cells. H2B-EGFP expressing ES cells were injected into C57BL/6 blastocysts. Chimeric mice were bred to 129/Tac mice for germline transmission. Resulting founder animals were bred with outbred ICR mice prior to arrival at The Jackson Laboratory. The mice were then backcrossed on to the C57BL/6J background for at least five generations.

| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||

| Molecular Note | The coding sequence of EGFP was placed C-terminally to the coding sequence of the human histone H2B gene. When placed under control of the chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer/promoter, a fluorescent fusion protein (H2B-EGFPis produced. Colony Maintenance
Breeding & Husbandry When maintaining a live colony, hemizygous mice are bred wildtype siblings. For mice on the STOCK background (Stock No. 005418), the donating investigator reports occasional silencing of the transgene when transmitted through the female germ line and recommends identifying mutant offspring by the fluorescent phenotype. | Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
