Mice homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) under the control of the chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer. Nucleosomes and chromatin in all cells fluoresce. Fluorescence is detectable during all phases of mitosis. The donating investigator reports occasional silencing of the transgene when transmitted through the female germline. This mutant mouse strain may be useful for in vivo subcellular high-resolution and 3-dimensional studies of dividing cells.In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Strain Development
A transgenic construct containing an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) was placed under the control of a chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer promoter. This construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-derived R1 embryonic stem (ES) cells. H2B-EGFP expressing ES cells were injected into C57BL/6 blastocysts. Chimeric mice were bred to 129/Tac mice for germline transmission. Resulting founder animals were bred with outbred ICR mice prior to arrival at The Jackson Laboratory. The mice were then backcrossed on to the C57BL/6J background for at least five generations.
