Strain Description
Hemizygous mice are viable and fertile with no gross or behavioral abnormalities. Flow cytometry identifies enhanced green fluorescent protein (EGFP) reporter expression in a pattern similar to the endogenous gene; in bone marrow, high expression is observed in myeloid cells, and to a lesser degree in B lymphocytes and erythroid cells. Expression in spleen is consistent with B cells and natural killer cells, while no expression is observed in thymus. These mice may be useful in studies of hematopoietic cell development, transcription factor pathways, and leukemia (including erythroleukemia).
Strain Development
A transgene was created to contain the -14 kb upstream regulatory element (URE), 2.1 kb promoter sequence, and cDNA sequence of the murine PU.1 (Sfpi1 gene) all positioned upstream of an internal ribosomal entry site-enhanced green fluorescent protein (IRES-EGFP, Clonetech) and followed by the rabbit beta-globin gene (containing exon 2-3) and polyadenylation signal. This transgene was injected into FVB/N fertilized oocytes and embryos were implanted into the uteri of pseudopregnant FVB/N foster mothers. Transgenic offspring (founder line 7) were bred to C57BL/6 mice for one generation. The resulting hemizygous mice (mixed C57BL/6;FVB/N background) were intercrossed for many generations prior to arrival at The Jackson Laboratory.

| | Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source.

A Sfpi1 mini-gene comprising a 3.4-kb genomic DNA fragment containing the -14-kb upstream regulatory element (URE), a 2.1-kb genomic segment containing the promoter, and a cDNA representing the processed transcript of the gene is followed by an internal ribosome entry site (IRES), the enhanced green fluorescent protein (EGFP) coding sequence, and exon 2, intron 2, exon 3 and the polyadenylation signal of the rabbit beta-globin gene. Cryorecovery - Standard. The recovery process begins when a signed agreement form is returned to the Customer Service Department after order placement. Although results vary by strain, at least two males and two females (two pairs) will be provided, typically within 15 weeks of our receipt of the signed agreement form. If the first recovery attempt is unsuccessful or only one pair is recovered, a second recovery will be done, extending the delivery time to approximately 25 weeks. At least one member of each pair will be of known genotype and will carry the mutation if it is a mutant strain. Please note that pairs may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation of the strain. Mating schemes are sometimes modified for successful cryopreservation. Price represents a repository maintenance fee, which includes the cost of recovery of the strain from the cryopreservation resource and the periodic replacement of the frozen embryos used for recovery.Cryorecovery to establish a Dedicated Supply for greater quantities of mice. One to two pairs will be recovered to establish a Dedicated Supply of mice. Price by quotation. The Jackson Laboratory's Genotype Promise
The Jackson Laboratory has rigorous genetic quality control and mutant gene
genotyping programs to ensure the genetic background of JAX
® Mice strains as
well as the genotypes of strains with identified molecular mutations.
JAX
® Mice strains are only made available to researchers after meeting our
standards. However, the phenotype of each strain may not be fully
characterized and/or captured in the strain data sheets.
Therefore, we
cannot guarantee a strain's phenotype will meet all expectations. To
ensure that JAX
® Mice will meet the needs of individual research projects
or when requesting a strain that is new to your research, we suggest ordering
and performing tests on a small number of mice to determine suitability for
your particular project.
