Type Mutant Stock; Targeted Mutation; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse Generation ? + F2p Donating Investigator W. Scott Young III, National Institute of Mental Health Description
Mice homozygous for this targeted mutation are viable, fertile, normal in size, exhibit apparently normal sexual behavior, and do not display any gross physical abnormalities. Homozygous mice exhibit less social agression, altered chemoinvestigatory behavior, and impaired social recognition. These mice may be useful in studies of agressive behavior, social motivation, and appropriate behavioral responses, and may be potential models of autism and agression accompanying dementia and traumatic brain injury.Development
A targeting vector was used to replace the coding region from before the initiating methionine to just upstream of the transmembrane VI region of the endogenous gene with a neomycin resistance cassette. The construct was electroporated into the 129X1/SvJ-derived RW-4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6 to generate mutant colonies on a mixed background.

Avpr1btm1Wsy/Avpr1btm1Wsy
involves: 129S4/SvJae * C57BL/6
- behavior/neurological phenotype
- abnormal social investigation (MGI Ref ID J:94939)
- no decrease in the amount of time mutant males spend sniffing a female during the second exposure period is seen unlike wild-type males
- decreased aggression (MGI Ref ID J:94939)
- males display a longer latency to attack and less agonistic behavior towards intruder males
- homeostasis/metabolism phenotype
- increased circulating testosterone level (MGI Ref ID J:94939)
- testosterone is elevated in young males (60-90 days old) but not in males older than 120 days

Colony Maintenance
Breeding & Husbandry When maintaining a live colony, these mice are bred as heterozygotes. Homozygous mice are viable and fertile with apparently normal sexual behavior, but the donating investigator does not attempt based on the possibility of behavior and parenting problems.
