Strain Description
Mice hemizygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express Enhanced Green Fluorescent Protein (EGFP) under the direction of the mouse Gad1 (glutamic acid decarboxylase 1 or GAD67) promoter. In the neocortex, EGFP expression is detected predominantly in layers 5B and 6, and to a lesser extent in layers 2/3. Low levels of fluorescence are detected in small cells with glial morphology. EGFP expression is also observed in area CA1 of the hippocampus, mainly in large interneurons in the oriens-alveus layers. In the neocortex of founder line 98 transgenic mice, fluorescence is observed in infragranular, calbindin immunoreactive interneurons with axonal arborizations in layer 1. In animals younger than two weeks of age, an increased number of EGFP labeled neurons is observed in the supragranular layers, and EGFP expression is also observed in some cells with pyramidal morphology. The fluorescently labeled populations of neurons in this founder line is distinct from the subset of somatostatin-containing (SOM+) interneurons in founder line 94 mice (Stock No. 006334), or in mice expressing Tg(GadGFP)45704Swn (Stock No. 003718), in location, neurochemical markers, axonal morphologies, and electrophysiological characteristics. This mutant mouse strain may be useful in studies of populations of inhibitory interneurons of the hippocampus and neocortex.
Strain Development
A transgenic construct containing EGFP gene under the control of the mouse Gad1, glutamic acid decarboxylase 1, promoter was injected into fertilized C57BL6 X SJL mouse eggs. Founder line 98 was established. Animals were initially bred to C57BL/6 mice and then bred to outbred CD-1 mice. The mice were then bred to outbred Hilltop Swiss ICR mice. The Donating Investigator maintained the strain by mating hemizygotes to outbred mice.

| | Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source.

Of 11 initial lines derived from founders shown by PCR analysis to carry the transgene, 5 that expressed EGFP in the brain continued to be maintained. Two of these, designated by the authors X94 and X98, in which cortical EGFP expression was limited to nonpyramidal cells were subjected to further analysis. |

The transgene comprises 10.3 kb of mouse genomic DNA containing the promoter region from the glutamic acid decarboxylase 1 (Gad1, commonly called GAD67) gene, including the 5' regulatory sequences, (noncoding) exon 1, intron 1 and a segment of exon 2, joined in-frame to the coding sequence of enhanced green fluorescent protein followed by an SV40 polyadenylation signal. The encoded fusion protein contains the first 17 amino acids of GAD1, 4 amino acids derived from the pEGFP-1 vector, and the entire enhanced green fluorescent protein. GFP expression is found in infragranular, calbindin-containing, layer 1-targeting cells that have propensity to fire low-threshold calcium spikes. [J:117601]

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |

or when requesting a strain that is new to your research, we suggest ordering
