Approximately half of the double homozygous targeted mutation mice die within days of birth. The surviving mice are subject to severe seizures that start in the second postnatal week then die within the following 2 weeks. Nissl staining of brain tissues revealed no structural abnormalities. No changes were found in the levels of selected synaptic proteins. Neurons experience sustained increases in Ca2+-dependent synaptic transmission when two or more action potentials are triggered in succession. Electrophysiological recordings from cultured hippocampal cells exhibit no detectable abnormalities. No protein products from the targeted genes are detected in brain tissue. This mutant mouse strain represents a model that may be useful in studies of synaptic vesicle trafficking in neurons.Development
Targeting vectors containing a neomycin resistance gene were used to replace exons 2-9 encoding residues 208-559 of Sv2a and exon 1 encoding residues 1-149 of Sv2b. The constructs were electroporated into 129P2/OlaHsd-derived E14.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. Chimeric animals were crossed with C57BL/6. The strain has been minimally backcrossed to C57BL/6 by the donating laboratory.

Breeding & Husbandry When maintaining a live colony, mice heterozygous for both mutations may be bred together. Once established, mice heterozygous for the Sv2a mutation and homozygous for Sv2b mutation may be bred together (or to wildtype mice). Mice homozygous for both mutations are not viable.

The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
