Generation DefinitionsDonating Investigator Dr. Thomas C. Sudhof, Stanford University School of Medicine Description
Mice homozygous for this targeted point mutation are viable and fertile and do not display any gross physical or behavioral abnormalities. The mutant gene is fully expressed. A two-fold decrease in the overall Ca2+ affinity of the protein and a corresponding decrease in the Ca2+ sensitivity of neurotransmitter release is observed. Nuclear Magnetic Resonance (NMR) spectroscopy shows no structural or conformational changes in the mutant protein. This mutant mouse strain represents a model that may be useful in studies of synaptic neurotransmitter release.Development
A targeting vector was used to create an R233Q substitution in the exon encoding residues 214-269 and insert a floxed neomycin resistance gene into the flanking intron. The construct was electroporated into 129P2/OlaHsd-derived E14.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. Chimeric animals were crossed with C57BL/6. The strain has been minimally backcrossed to C57BL/6 by the donating laboratory.

involves: 129P2/OlaHsd * C57BL/6
- nervous system phenotype
- abnormal excitatory postsynaptic currents
- the ESPC amplitudes of mutant neurons were smaller than controls during repetitive stimulation; facilitation of the response was seen at various frequencies whereas synaptic depression was seen in controls (MGI Ref ID J:76452)
- abnormal neurotransmitter secretion
- the fast, synchronous, Ca2+ dependent phase of neurotransmitter release is affected, while the spontaneous release of neurotransmitters is unaffected (MGI Ref ID J:76452)
- the Ca2+ dependent synaptic release probability was decreased two-fold, but was not due to a decrease in the readily releasable pool of neurotransmitters (MGI Ref ID J:76452)

| Molecular Note | Insertion of a neomycin resistance cassette into intron 1 and an arginine to glutamine point mutation at position 233 of exon 1 (R233Q) was introduced via homologous recombination. Western blot of brain homogenates from homozygous mutant animals verifiedthe presence of protein expression and enzyme assays demonstrated similar phospholipid- and syntaxin-binding ability compared to wild-type. However, calcium affinity was decreased by 2-fold. [MGI Ref ID J:76452] | ||

Supply Notes
- Cryorecovery - Standard.
Progeny testing is not required.
The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
Mice recovered can be used to establish a dedicated colony to contractually supply you mice according to your requirements. Price by quotation.
