Hemizygous mice are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. Transgene expression is observed in all areas that endogenously express Sim1, including paraventricular hypothalamus and other parts of the brain. When these Sim1-Cre mice are bred with mice containing a loxP-flanked sequence of interest, Cre-mediated recombination will result in deletion of the flanked sequences in Sim1-expressing tissues (including hypothalamus). As such, Sim1-Cre transgenic mice may be useful in studying body weight homeostasis, obesity, leptin metabolism, or as a reporter strain for Sim1-transcription factor activity.In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
Development
A bacterial artificial chromosome (BAC) was modified by inserting a Cre recombinase sequence into the translational start site of the mouse Sim1 gene (removing the first 31 basepairs of Sim1. Correctly transformed and targeted bacteria had the selection cassette transiently removed (leaving a single frt site downstream of the Cre recombinase). The resulting construct, called "Sim1-Cre," was used for pro-nuclear injection into fertilized one-cell stage FVB embryos. Transgenic offspring (founder line 1) were established and maintained on a mixed genetic background (C57BL/6, 129/SvJ, and FVB) prior to arrival at The Jackson Laboratory (as Stock No. 006395). Upon arrival, mice were backcrossed to C57BL/6J for at least 5 generations to establish this congenic strain.
