Type JAX® GEMM® Strain - Coisogenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Transgenic Mating System +/+ sibling x Hemizygote (Female x Male) Species laboratory mouse Donating Investigator Brian Soper, The Jackson Laboratory Generation ?+F2 (20-DEC-07) Strain Description
This transgenic mouse line with an "enhanced" GFP (EGFP) cDNA under the control of a chicken beta-actin promoter and cytomegalovirus enhancer makes all of the tissues, with the exception of erythrocytes and hair, appear green under excitation light. The viability of homozygotes is unknown.Strain Development
The EcoRI sites included in the PCR primers were used to introduce the amplified EGFP cDNA into a pCAGGS expression vector containing the chicken beta-actin promoter and cytomegalovirus enhancer, beta-actin intron and bovine globin poly-adenylation signal. The entire insert with the promoter and coding sequence was excised with Bam-HI and SalI and gel-purified. The transgenic construct was was introduced into C57BL/6 donor eggs. Founder mice were bred to C57BL/6 mice, and backcrossed to the same for 11 generations.

| Expressed Gene | GFP, Green Fluorescent Protein, jellyfish | ||
| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||
| Promoter | ACTB, actin, beta, human | ||
| General Note | This transgene was made using the same construct as Tg(ACTB-EGFP)1Osb, but in this case it was injected into C57BL/6 fertilized oocytes. According to the Center for Animal Resources and Development Database (CARD R-Base), which distributes it, the transgene integrated on Chr 14D1. | ||
| Molecular Note | The transgene comprises the cDNA encoding enhanced green fluorescent protein (EGFP), from the jellyfish Aequoria victoria, downstream of the "CAG promoter" -- which consists of the cytomegalovirus immediate early (CMV-EI) enhancer followed by a 1.3-kb DNA segment including the promoter, first exon and first intron of the chicken beta-actin gene, with the 3' splice junction sequence replaced by that of the rabbit hemoglobin beta gene -- and followed by the rabbit beta-hemoglobin polyadenylation signal and 3' flanking sequence. | Breeding & Husbandry | When maintaining a live colony, these mice are bred as hemizygotes. The Donating Investigator maintained the line using a hemizygote x hemizygote breeding scheme and has not tried to make the strain homozygous. |

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
