Strain Description
The coding region and part of the 3' non-translated region of the Omp gene was replaced by taulacZ, a fusion protein of the microtubule-binding protein tau (from bovine origin) and beta-galactosidase. The targeted mutation results in a knockout. Mature olfactory sensory neurons express beta-galactosidase at high levels. These neurons can be revealed by histochemistry of beta-galactosidase enzymatic activity, or by immunofluorescence with anti-beta-galactosidase antibodies. Homozygous mice are subfertile.

Strain Development
The Omptm1Mom targeted mutation was generated in 129S2/SvPas-derived D3 ES cells by replacing the single Omp coding exon with tau-lacZ followed by an LTNL cassette. After expanding targeted clone# B38 the LTNL was excised by transiently transfecting with a plasmid encoding CMV-cre recombinase and these cells were negatively selected for recombination using ganciclovir. Clone B38/CreA5, which had the LTNL excised, was injected into C57BL/6 blastocysts and the resulting strain was interbred to homozygosity on this mixed B6;129S2 background.

Molecular Note Insertion of a tau-lacZ-loxP-tk-neo-loxP expression cassette deleted the coding exon and placed the tau-lacZ sequences under control of the endogenous regulatory sequences. Transient expression of Cre recombinase in C57BL/6 blastocysts excised the thymidine kinase and neomycin sequences. Expression of the tau-lacZ fusion protein in heterozygous mutant mice was detected in olfactory axon termini in individual glomeruli. [MGI Ref ID J:36595]
