Hemizygous transgenic mice are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express the mouse transducin-like enhancer of split 1, homolog of Drosophila E(spl) gene, Tle1 (also known as Groucho-related gene 1; Grg1) under the regulation of the chicken beta actin promoter (ACTB) and the cytomegalovirus (CMV) intermediate-early enhancer. An internal ribosome entry sequence (IRES) followed by the human alkaline phosphatase (ALPP) reporter gene inserted downstream of the Tle1 gene directs co-expression of ALPP and TLE1. TLE1 and ALPP co-expression is blocked, however, by the presence of a loxP-flanked Bgeo (beta-galactosidase/neomycin resistance fusion protein) cassette inserted between the ACTB/CMV promoter and the Tle1 coding sequence. In the absence of Cre recombinase, beta-galactosidase activity is detected in all tissues tested including pancreas, lung, kidney, cerebellum, heart, muscle and lens and retina. Diminished beta-galactosidase activity is observed in liver tissue. When transgenic mice are crossed with a Cre recombinase-expressing strain, lacZ expression in the resulting offspring is replaced by both ALPP and TLE1 protein in cre-expressing tissues. These mice may be useful in studying lung tumorigenesis.Strain Development
A transgenic construct containing the human alkaline phosphatase, placental (Regan isozyme), gene ALPP, the mouse transducin-like enhancer of split 1, homolog of Drosophila E(spl) gene Tle1, loxP site flanked Bgeo reporter (beta-galactosidase/neomycin resistance fusion gene)/SV40 polyadenylation signal segment and IRES (Internal Ribosomal Entry) sequence under the control of the a chicken beta actin promoter coupled with the cytomegalovirus (CMV) immediate early enhancer, was introduced into R1 embryonic stem (ES) cells. ES cell clones containing a single copy of the construct were used to generate aggregated chimeric mice. Resulting male chimeric animals were bred with CD1 female mice.
