Type JAX® GEMM® Strain - Coisogenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Transgenic Mating System +/+ sibling x Hemizygote (Female x Male) Species laboratory mouse Donating Investigator Tibor Kristian, University of Maryland Generation N7+ (09-JUL-07) Strain Description
Hemizygotes are viable and fertile. These transgenic mice express a mitochondrial-specific EYFP fusion protein under the control of a tetracycline-responsive promoter element (TRE;tetO). When hemizygotes are bred with another transgenic mouse expressing either reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) under the regulation of tissue-specific promoter, tissue-specific expression of EYFP in mitochondria of the bitransgenic offspring can be regulated with the tetracycline analog, doxycycline.When bred to a strain expressing rtTA or tTA in forebrain neurons (see Stock No. 003010 for example), this mutant mouse strain may be useful in studies of neuronal mitochondrial dysfunction.
Strain Development
A transgenic fusion protein construct containing the mitochondrial targeting sequence from the human COX8 gene and Enhanced Yellow Fluorescent Protein (EYFP) was placed downstream of a tet-responsive promoter element (TRE) and upstream of a beta-globin poly A signal. The construct was injected into fertilized C57BL/6 eggs. Founder animals were bred to C57BL/6 for six generations by the donating investigator.

| Yellow Fluorescent Protein (YFP) is a derivative of Green Fluorescent Protein (GFP), a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance fluorescence intensity and shift the wavelength emitted when excited. When YFP is utilized in a transgenic construct, tissue expressing sufficient amounts of YFP will fluoresce yellowish-green when exposed to a 513 nm light source. | |||
| Expressed Gene | COX8A, cytochrome c oxidase subunit 8A (ubiquitous), human | ||
| Promoter | tetO, tet operator, | ||
| Molecular Note | A construct encoding a fusion protein comprising the mitochondrial targeting sequence from human cytochrome c oxidase subunit 8A (ubiquitous) joined to enhanced yellow fluorescent protein (EYFP) was placed downstream of the tetracycline responsive element (tetO) and upsteam of a beta-globin polyadenylation signal. A mouse with this transgene can be bred with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) under control of a selected promoter to produce bitransgenic mice in which mitochondrial EYFP expression can be regulated in a tissue-specific manner by administration/removal of the tetracycline analog, doxycycline. | Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
