Former Names B6;129P2-Olfr151tm13(rI7)Mom/MomJ (Changed: 08-FEB-07 ) Type Mutant Stock; Targeted Mutation; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Species laboratory mouse Donating Investigator Peter Mombaerts, Max Planck Institute of Biophysics Description
The coding region of the Olfr151 olfactory receptor was replaced by that of the rat I7 olfactory receptor. Olfactory sensory neurons that express the mutated locus co-express both GFP and taulacZ by virtue of IRES-mediated co-translation. These neurons can be revealed by the intrinsic fluorescence of GFP, by immunofluorescence with anti-GFP antibodies, by histochemistry of beta-galactosidase enzymatic activity, or by immunofluorescence with anti-beta-galactosidase antibodies. Expression of the inserted rat sequence instead of the endogenous sequence causes the location of axonal convergence in expressing neurons to be rerouted tod a more anterior, ventral position. These neurons respond to octanal.Development
The Olfr151tm13(rI7)Mom mutation was generated in 129P2/OlaHsd derived E14 ES cells. To excise the neomycin resistance sequence of the LNL cassette, a resulting mouse was bred with a mouse carrying Tg(EIIa-cre)Lmgd, a transgene generated in FVB/N but having been backcrossed at least 10 times to C57BL/6J before this cross. The transgene was then bred out of the line and the mice were intercrossed to homozygosity. The plasmid that was used to generate this mutation is banked and described at Addgene.
