Type Mutant Stock; Targeted Mutation; Transgenic; Additional information on Genetically Engineered and Mutant Mice. Visit our online Nomenclature tutorial. Species laboratory mouse Donating Investigator Peter Mombaerts, The Rockefeller University Development
A taulacZ cassette was inserted downstream of a minigene of 9.2 kb of genomic sequence that included the coding and regulatory sequence Olfr151 and the construct was microinjected into (CBA x C57BL/6J)F2 pronuclei. Several founders were obtained and the lines were bred to a strain bearing Olfr160tm1Mom (commonly referred to as M71->M72-IRES-tauGFP), which was generated in 129P2/OlaHsd-derived E14 ES cells and outcrossed to C57BL/6J. In 2007 sperm was cryopreserved from males carrying both the transgene and the targeted mutation.

| Molecular Note | The entire coding region of Olfr160 was replaced with Olfr151 and an IRES-tauGFP gene introduced via homologous recombination. The lacZ gene was fused after the stop codon and co-expressed with the mutant protein. [MGI Ref ID J:90716] | ||

| Site of Expression | GFP is expressed in olfactory sensory neurons that express the olfactory receptor Olfr151. LacZ is expressed in Zone 4 of the olfactory epithelium. | ||
| Expressed Gene | GFP, Green Fluorescent Protein, jellyfish | ||
| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||
| Promoter | Olfr151, olfactory receptor 151, mouse, laboratory | ||
| Molecular Note | This transgene, generated through pronuclear injection, has 9.2 kb of genomic sequence, with 2.3 kb upstream of the putative transcription start site and 3.8 kb downstream of the coding region, and a taulacZ cassette inserted downstream of the coding sequence.
