Strain Description
Mice hemizygous for this Cre-conditional TEL-AML1 (or iZ/EG-TEL-AML1) transgene are viable and fertile. Homozygotes die in utero, presumably due to high lacZ expression. Prior to Cre-mediated excision of the "floxed" STOP sequence, high expression of lacZ is observed in cells and tissues. When bred to Cre recombinase transgenic mice, the STOP sequence (and beta-geo) is removed in the resulting offspring, allowing transcription/co-expression of both the human TEL-AML1 fusion protein and EGFP in all cre-expressing cells. TEL-AML1 transcripts are not observed in adult organ tissues prior to excision of the floxed sequences. Following Cre-mediated deletion of the STOP sequence (by B6.Cg-Tg(Tek-cre)12Flv/J, Stock No. 004128), Western blot analysis reveals that EGFP levels are well correlated with TEL-AML1 transcript levels. While global expression of TEL-AML1 leads to embryonic lethality (E7.5), hematopoietic and endothelial expression leads to malignancy in B- and T- lymphoid cells and secondary mutations that closely resemble human TEL-AML1 associated leukemia. These transgenic mice may be useful for global expression of lacZ or, when crossed to a cre-expressing strain, for studying hematopoietic development, cancer, and leukemia.
Strain Development
The iZ/EG-TEL-AML1 transgenic construct contains the CMV enhancer/chicken beta-actin promoter, a loxP-flanked STOP sequence (beta-geo reporter followed by SV40 polyadenylation signal), the human TEL-AML1 fusion protein sequence (coding for the translocation t(12;21)(p13;q22)), an internal ribosomal entry site, and an enhanced green fluorescent protein (EGFP) gene. This construct was electroporated into 129X1/SvJ x 129S1/Sv-derived R1 embryonic stem (ES) cells. ES cells with strong lacZ expression, single copy transgene integration, and efficient expression of EGFP following Cre-mediated excision of the STOP sequence were selected and aggregated with E2.5 8-cell CD1 mouse embryos. Following transfer into pseudo-pregnant females, chimeric males were mated to wildtype CD1 females to establish founder line A6. This strain was maintained by breeding hemizygous mice to CD1 or wildtype siblings for approximately six generations prior to arrival at The Jackson Laboratory. Upon arrival, these mice were bred at least one generation to C57BL/6J.

| | Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. The original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source.

The Jackson Laboratory has rigorous genetic quality control and mutant gene
genotyping programs to ensure the genetic background of JAX
® Mice strains as
well as the genotypes of strains with identified molecular mutations.
JAX
® Mice strains are only made available to researchers after meeting our
standards. However, the phenotype of each strain may not be fully
characterized and/or captured in the strain data sheets.
Therefore, we
cannot guarantee a strain's phenotype will meet all expectations. To
ensure that JAX
® Mice will meet the needs of individual research projects
or when requesting a strain that is new to your research, we suggest ordering
and performing tests on a small number of mice to determine suitability for
your particular project.
