Type JAX® GEMM® Strain - Congenic Additional information on JAX® GEMM® Strains. Type JAX® GEMM® Strain - Mutant Strain Type JAX® GEMM® Strain - Targeted Mutation Mating System Heterozygote x Homozygote (Female x Male) Species laboratory mouse Donating Investigator Yuqing Chen, University of Michigan, Medical School Generation N?F0 (03-DEC-07) Strain Description
Mice homozygous for this SM22alpha-CreKI allele are viable and fertile. These mice have a Cre-recombinase gene inserted into the endogenous transgelin (SM22alpha) locus. The donating investigator reports that this mutation results in a loss of function of the targeted gene. Cre recombinase activity is shown in adult smooth muscle cells (such as arteries, veins, and visceral organs) and cardiac myocytes, but activity is not observed in the same embryonic tissues. These SM22alpha-CreKI mice may be useful for Cre-lox technology applications in studying smooth muscle and cardiac gene function, as well as cardiovascular disease.Strain Development
A targeting vector was designed to insert a Cre-recombinase coding sequence (followed by a PGK-neo cassette) in-frame at the SM22alpha (Tagln or transgelin) initiation site. The construct was electroporated into 129S6/SvEvTac-derived "CMT-1" embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. Chimeric animals were crossed with C57BL/6 to generate heterozygotes. This strain was backcrossed to C57BL/6J for at least 12 generations prior to arrival at The Jackson Laboratory. The Y chromosome may not have been fixed to the C57BL/6J genetic background.

| Expressed Gene | cre, cre recombinase, bacteriophage P1 | ||
| Cre recombinase is an enzyme derived from the bacteriophage P1 that specifically recognizes loxP sites. Cre has been shown to effectively mediate the excision of DNA located between loxP sites. After the excision event, the DNA ends recombine leaving a single loxP site in place of the intervening sequence. | |||

| Standard Supply | Repository-Live. A collection of over 1000 strains maintained as live colonies. Individual colonies are sized to meet current customer demand. Delivery for orders of 10 mice or less ranges on average from one to eight weeks; mice are generally shipped between four to six weeks of age with a maximum shipping age of ~nine weeks. Colony sizes do not generally support stringent age specifications for large volumes of mice; however custom orders and larger quantities of mice are easily arranged. Estimated ship dates for all orders provided within 48 hours of order placement. |
