Mice homozygous for this targeted mutation are viable and fertile and do not display any gross physical or behavioral abnormalities. There is no identifiable phenotype in these mice. Expression is eliminated in the brain as determined by RT-PCR and Northern analyses, and in situ hybridization and immunofluorescence of spinal cord and retina. This strain may be helpful in studies to further delineate the role of this gene.Development
An FRT-flanked PGK-neomycin drug resistance cassette was targeted to replace a 2.3 kb genomic segment containing exons 6 and 7 in a mouse Glra2-containing BAC. A NotI fragment from the BAC was electroporated into an 129S4/SvJae-derived J1 embryonic stem (ES) cell line. Homologously recombined ES cells were injected into blastocysts to generate chimeric animals. The foundation line was backcrossed to C57BL/6.

| Molecular Note | A vector was designed to replace exons 6 and 7 with an FRT-flanked PGK-neomycin cassette. These exons encode two transmembrane domains and a large portion of its ligand binding domain. Alternative splicing of exon 5 with exon 8 would result in an immediate premature stop codon and should lead to a nonsense-mediated degradation of the mRNA. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. The total number of animals provided, their gender and genotype will vary. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 13 and 16 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility.
