Liver-specific expression of the transgene is exhibited in this strain of mice as determined by RT-PCR. When crossed with a F5 targeted mouse (Stock No. 004078), homozygous knockout/hemizygous transgenic mice are grossly normal and show no evidence of spontaneous hemorrhage; compound mutants have a plasma F5 level that is ~45% that of wildtype mice, no platelet F5, slightly prolonged bleeding times, and show transgene rescue of both mid-embryonic lethality and perinatal hemorrhage phenotypes associated with F5 knockouts.Development
A mouse Alb (albumin) promoter drives expression of the mouse F5 (FV) transgene including the entire coding sequence. An internal ribosome entry site sequence (IRES) followed by an enhanced green fluorescent protein (EGFP) coding sequence and a bacterial kanamycin resistant gene (KAN) were also built into the cassette. A chimeric intron with optimized splice signals from the pCI expression vector was also introduced upstream of the F5 cDNA in the transgene cassette to enhance gene expression. No EGFP expression is observed. The vector was injected into fertilized oocytes created by mating B6SJLF1 or C57BL/6 female mice with B6SJLF1 male mice. This line has been backcrossed minimally (perhaps twice) to C57BL/6 by the donating laboratory.
