Type Consomic; Additional information on Consomic Mice. Type Mutant Strain; Transgenic; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse and M. m. domesticus poschiavinus Donating Investigator Eva Eicher, The Jackson Laboratory Development
Tg(Sry-EGFP)92Ei was generated via random insertion after micro-injection into the pronucleus of C57BL/6J fertilized eggs and resulted in stock #003927, C57BL/6J-Tg(Sry-EGFP)92Ei/EiJ. Tg(Sry)4Ei was also generated via random insertion after micro-injection into the male pronucleus of fertilized eggs from C57BL/6J females bred with C57BL/6J-Chr YPOS hermaphrodites. These strains were bred together to produce this strain and sperm was cryopreserved from a male homozygous for Tg(Sry-EGFP)92Ei and carrying both Tg(Sry)4Ei and Chr YPOS.

assigned by genotype
The following phenotype information may relate to a genetic background differing from this JAX® Mice strain.
Tg(Sry)4Ei/0
C57BL/6JEi-Tg(Sry)4Ei Chr YPOS/EiJ
- reproductive system phenotype
- primary sex reversal (MGI Ref ID J:31072)
- chromosomal females carrying this transgene develop as males
- chromosomal males that would have developed as sex-reversed females, due to the m. poschiavinus Y chromosome, develop as males when carrying this transgene
Tg(Sry-EGFP)92Ei/0
C57BL/6J-Tg(Sry-EGFP)92Ei
- normal phenotype
- no abnormal phenotype detected (MGI Ref ID J:73280)
- mice have no overt abnormal phenotype; marker expression is spatially similar to endogenous Sry expression

| Allele Symbol | Tg(Sry)4Ei | ||
|---|---|---|---|
| Allele Name | transgene insertion 4, Eva M Eicher | ||
| Allele Type | Transgenic (random, expressed) | ||
| Strain of Origin | C57BL/6J-Chr YPOS | ||
| Site of Expression | Fluorescence is detected in pre-sertoli and pre-granulosa cells of the genital ridge, expression is stronger in homozygotes. Expression is also detected in tyrosine hydroxylase-expressing neurons of the substantia nigra of the midbrain and the medial mammillary bodies of the hypothalamus in males but not females. | ||
| General Note | The single founder male carried two independent multiple transgene insertions designated Tg(Sry)4Ei (with a high copy number) and Tg(Sry)5Ei (with a low copy number). Only transgenics carrying Tg(Sry)4Ei alone were used in subsequent experiments. | ||
| Molecular Note | This transgene consists of an inbred 129 strain derived 14.6 Kb genomic fragment containing the entire Sry open reading frame and no other genes. [MGI Ref ID J:31072] | ||
| Allele Symbol | Tg(Sry-EGFP)92Ei | ||

| Allele Type | Transgenic (Reporter) | ||
| Common Name(s) | Tg92Ei; | ||
| Strain of Origin | C57BL/6J | ||

| Expressed Gene | GFP, Green Fluorescent Protein, jellyfish | ||
| Green Fluorescent Protein (GFP), derived from the jellyfish Aequorea victoria, is a versatile reporter molecule which has found use in many biological applications. In some constructs the original molecule has been modified in order to enhance its fluorescence intensity (EGFP, enhanced GFP). When utilized in a transgenic construct, tissue expressing sufficient amounts of GFP will fluoresce when exposed to a 488 nm light source. | |||
| Promoter | Sry, sex determining region of Chr Y, mouse, laboratory | ||
| Molecular Note | The transgenic vector was composed of the Sry promoter region from bp 542-8304 inserted upstream of the EGFP-1 coding region. The construct contains two SV40 early mRNA polyadenylation signal in place of the endogenous Sry 3' UTR. Expression of EGFP indicates that the spatial expression of EGFP mRNA and protein is similar to endogenous Sry . [MGI Ref ID J:73280] | ||
