Generation DefinitionsDonating Investigator David L. Garbers, U.T. Southwestern Medical Center Description
Homozygotes are viable. Males, but not females, are completely infertile. The targeted mutation fails to significantly alter sperm production, protein tyrosine phosphorylation associated with capacitation, induction of the acrosome reaction, forward velocity or percentage of motility. Sperm cells fail to acquire hyperactivated motility associated with penetration of the extracellular matrix of the egg. SDS/PAGE and immunoblotting of total sperm protein were used to confirm the absence of gene product. This strain may be useful in studies of male fertility.Development
A targeting vector was designed to replace exons 2-5 of the gene with a neomycin drug resistance cassette. The vector was electroporated into 129S6/SvEvTac-derived SM1 embryonic stem (ES) cells. Chimeric mice were generated and backcrossed once to C57BL/6 to produce heterozygotes.

- reproductive system phenotype
- asthenozoospermia
- although mutant sperm exhibit normal forward velocity and percentage of motile cells, they are unable to generate the hyperactivated form of sperm motility required for fertilization, as revealed by the absence of high track velocity and nonlinear trajectory spermatozoa in mutant sperm cell populations (MGI Ref ID J:86972)
- in mutant sperm, normal forward velocity is maintained despite a slight reduction in average flagellar amplitude and a small increase in the rate of flagellar beat (MGI Ref ID J:86972)
- in a high-viscosity medium, mutant sperm lose their ability to swim forward, whereas wild-type sperm remain progressively motile with reductions in forward velocity of ~50% (MGI Ref ID J:86972)
- impaired acrosome reaction
- mutant sperm are unable to penetrate the zona pellucida, despite normal forward velocity and percentage of motile cells (MGI Ref ID J:86972)
- however, mutant sperm exhibit normal protein tyrosine phosphorylation associated with capacitation, and initiate a normal acrosome reaction in response to intact zona pellucida (MGI Ref ID J:86972)
- impaired fertilization
- male infertility
- male, but not female, homozygotes are incompletely infertile under normal breeding and in vitro conditions (MGI Ref ID J:86972)
- however, mutant males display no significant differences in body and testis weights, testis and sperm morphology or epididymal sperm counts relative to wild-type littermates (MGI Ref ID J:86972)
The following phenotype information may relate to a genetic background differing from this JAX® Mice strain.
Catsper2tm1Gar/Catsper2tm1Gar
involves: 129S6/SvEvTac
- reproductive system phenotype
- abnormal sperm physiology
- calcium-selective pH sensitive sperm current (ICatSper) is not detectable (MGI Ref ID J:116406)

| Molecular Note | A neomycin selection cassette replaced a genomic fragment encompassing exons 2 through 5. The deleted region included the start codon and encoded the first three transmembrane helices. Protein was undetected in homozygous mutant mice by Western blot and immunocytochemical analyses of spermatozoa. [MGI Ref ID J:86972] | ||

Progeny testing is not required.
The average number of mice provided from recovery of our cryopreserved strains is 10. The total number of animals provided, their gender and genotype will vary. We will fulfill your order by providing at least two pair of mice, at least one animal of each pair carrying the mutation of interest. Please inquire if larger numbers of animals with specific genotype and genders are needed. Animals typically ship between 11 and 14 weeks from the date of your order. If a second cryorecovery is needed in order to provide the minimum number of animals, animals will ship within 25 weeks. IMPORTANT NOTE: The genotypes of animals provided may not reflect the mating scheme utilized by The Jackson Laboratory prior to cryopreservation, or that discussed in the strain description. Please inquire about possible genotypes which will be recovered for this specific strain. The Jackson Laboratory cannot guarantee the reproductive success of mice shipped to your facility. If the mice are lost after the first three days (post-arrival) or do not produce progeny at your facility, a new order and fee will be necessary.Cryorecovery to establish a Dedicated Supply for greater quantities of mice.
Mice recovered can be used to establish a dedicated colony to contractually supply you mice according to your requirements. Price by quotation.
