Type Mutant Stock; Targeted Mutation; Additional information on Genetically Engineered Mutant Mice. Species laboratory mouse Donating Investigator David Garbers, U.T. Southwestern Medical Center Description
Homozygotes are viable. Males, but not females, are completely infertile. The targeted mutation fails to significantly alter sperm production, protein tyrosine phosphorylation associated with capacitation, induction of the acrosome reaction, forward velocity or percentage of motility. Sperm cells fail to acquire hyperactivated motility associated with penetration of the extracellular matrix of the egg. SDS/PAGE and immunoblotting of total sperm protein were used to confirm the absence of gene product. This strain may be useful in studies of male fertility.Development
A targeting vector was designed to replace exons 2-5 of the gene with a neomycin drug resistance cassette. The vector was electroporated into 129S6/SvEvTac-derived SM1 embryonic stem (ES) cells. Chimeric mice were generated and backcrossed once to C57BL/6 to produce heterozygotes.

involves: 129S6/SvEvTac * C57BL/6J
- reproductive system phenotype
- asthenozoospermia (MGI Ref ID J:86972)
- though sperm exhibited normal forward velocity and percentage of motile cells, sperm was unable to generate the hyperactive form of motility necessary for fertilization
- male infertility (MGI Ref ID J:86972)
The following phenotype information may relate to a genetic background differing from this JAX® Mice strain.

| Molecular Note | A neomycin selection cassette replaced a genomic fragment encompassing exons 2 through 5. The deleted region included the start codon and encoded the first three transmembrane helices. Protein was undetected in homozygous mutant mice by Western blot and immunocytochemical analyses of spermatozoa. [MGI Ref ID J:86972] | ||
