A targeting construct with the SA-PhiC310pA-PGKneopA cassette, which contains the mouse codon-optimized site-specific recombinase PhiC31o sequence (with a C-terminal SV40 nuclear localization signal) driven by the phosphoglycerate kinase 1 (PGK) promoter, and bovine growth hormone polyadenylation (bpA) sequence was inserted into the Gt(ROSA)26Sor locus via electroporation of 129S4/SvJaeSor-derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were identified and used to generate mutant mice. Homozygous mice were maintained on a 129S4/SvJaeSor background prior to arrival at The Jackson Laboratory. The mice were then crossed to C57BL/6J (Stock No. 000664) for one generation.
